Analysis of associations of polymorphous markers ofTCF7L2 gene with diabetes mellitus of the 2nd type in the case of residents of the Republic of Tatarstan

The aim of the study was to investigate the possible association of the IVS3 С>Т
(rs7903146) and IVS4 G>Т (rs12255372) polymorphisms of the transcription factor 7-like
2 (TCF7L2) gene with type 2 diabetes mellitus (T2DM) in the Republic of Tatarstan (RT).
Materials and Methods. The study involved 76 adult patients (30% men and 70% women; 38-77 years old)
with type 2 diabetes mellitus and overweight or obesity. The distribution of genotypes and alleles of the
rs7903146 TCF7L2 gene polymorphism was compared with the literature data, for the rs12255372 TCF7L2
gene polymorphism was used our own sample (people without T2DM). All patients underwent clinical
and laboratory examination. Results. In the analysis of the frequency distribution of alleles and genotypes
of polymorphic markers rs7903146 and rs12255372 of the TCF7L2 gene in the study and control groups
statistically significant differences were not found. Correlation was found between carriers of genotype TT
and allele T of polymorphic marker rs7903146 of the TCF7L2 gene and the level of triglycerides (p=0,04),
increased average waist-hip ratio(p=0.005). Among patients with T2DM in the group withcarriers of allele
T of the rs 12255372 TCF7L2 gene was observed a significant increase average waist-hip ratio compared
with GG genotype (p=0,04). The connection between the carriers of genotype TT and allele T with such
parameters as BMI, total cholesterol, HDL and low density have not been identified.Conclusions. There was
a positive correlation of the risk alleles of the TCF7L2 gene with signs of obesity (the waist-hip ratio) and
metabolic disorders. In the analysis of the frequency distribution of alleles and genotypes of two polymorphic
markers TCF7L2 gene in the study and control groups was not found statistically significant differences. To
obtain more objective picture of distribution of risk alleles, further studies are needed with the expansion of the
volume of sample among patients and healthy individuals.
